Expression of Th1 and Th2 Cytokine and Associated Transcription Factors in Peripheral Blood Mononuclear Cells and Correlation with Disease Severity
Authors
Abstract:
Background: Psoriasis is a T cell-mediated autoimmune disease with elevated level of pro-inflammatory cytokines belonging mainly to Th1 pathway. We investigated whether treatment with micronutrients along with methotrexate (MTX) is able to modulate mRNA expression of Th1 and Th2 patterns and its correlation with disease severity. Methods: Thirty plaque type psoriasis patients with Psoriasis Area and Severity Index (PASI) higher than 10 recruited; 15 non- micronutrients taker (NMT) patients, treated by MTX daily (0.2-0.3 mg/kg/week) and 15 micronutrients taker (MT) patients treated by MTX plus micronutrient supplement daily for 12 weeks. Blood samples collected at baseline and after 12 weeks. Taqman quantitative real-time polymerase chain reaction was applied to analyses the expression of Th1 (T-bet, IL-12, IFN- γ) and Th2 (GATA-3, IL-4) pathway. Disease severity measured under PASI scoring system. Results: Significant clinical improvement in MT group was accordance with significant down-regulation of Th1 and up-regulation of Th2 studied markers (P<0.05). Respect to PASI-75 cut-point, expression of IFN-γ in MT group with upper PASI-75 was significantly lower than in related patients in NMT group (P=0.05). Also mRNA expressions of GATA3 and IL-4 in MT group with upper PASI-75 were significantly higher than patients in NMT group respectively (P=0.05, P=0.04) Conclusion: According to significant attenuating of PASI score correlated with upregulation of Th2 pathway in favor of MT group, consumption of micronutrients in combination MTX in psoriasis patients are suggested. Our results contribute to a better understanding of methotrexate immune-pathogenesis mechanisms and its correlation to clinical response in psoriasis.

Different Expressions of Specific Transcription Factors of Th1 (T-bet) and Th2 cells (GATA-3) by Peripheral Blood Mononuclear Cells From Patients With Multiple Sclerosis
Introduction: Multiple Sclerosis (MS) is an inflammatory disorder caused by self-reactive Th1 lymphocytes, while Th2 cells may confer protection. The Th1 and Th2 cell differentiation are regulated by specific transcription factors, especially T-bet and GATA-3, respectively. This investigation aimed to measure the T-bet and GATA-3 expression by Peripheral Blood Mononuclear Cells (PBMCs) obtained...
full textTh1 and Th2 Cytokine Profiles in Malignant Pleural Effusion
Background: The alteration of Th1 and Th2 cytokine levels is the subject of controversy in pleural effusions caused by malignancy, a situation that favors a Th2 immune response. Objective: To examine the different levels of IL-4 and IL-10 (Th2 cytokines), and IL-2 and interferon-γ (IFN-γ) (Th1 cytokines) in malignant and non-malignant pleural effusions. Method: The cytokine levels in pleural fl...
full textProliferation and TH1/TH2 Cytokine Production in Human Peripheral Blood Mononuclear Cells after Treatment with Cypermethrin and Mancozeb In Vitro
In recent times, human cell-based assays are gaining attention in assessments of immunomodulatory effects of chemicals. In the study here, the possible effects of cypermethrin and mancozeb on lymphocyte proliferation and proinflammatory (tumor necrosis factor (TNF-) α) and immunoregulatory cytokine (interferon- (IFN-) γ, interleukins (IL) 2, 4, 6, and 10) formation in vitro were investigated. H...

Background: Pre-eclampsia (PE) is known as a main factor contributing to fetomaternal mortality, which might affect 2-8% of all pregnancies after the twentieth week of gestation. The balance of T helper subsets is essential to sustain a normal pregnancy and preventing fetomaternal complications. Objective: To investigate differences in the levels of transcription factors and cytokine gene e...

Background: Multiple sclerosis (MS) is a T cell mediated autoimmune disease with unknown etiology. Appropriate MS therapeutic strategies need thorough understanding of both disease etiology and pathogenesis mechanisms. Ligation of TLR-2 and TLR-4 stimulates the production of several cytokines leading to CNS autoimmunity and neurodegenerative diseases. Objective: To find a relationship between M...
